Daconib (Dacomitinib 45 mg tablet)

Daconib (Dacomitinib 45 mg tablet) is an oral small-molecule kinase inhibitor. It was approved by the FDA on September 27, 2018, for the first-line treatment of non-small cell lung cancer in adult patients with epithelial growth factor receptor (EGFR) exon 19 deletion mutation or exon 21 L858R substitution.

What is the dose of Dacomatinib Tablet?

• The recommended dose is 45 mg orally once daily with or without food.
• In certain adverse reactions, the recommended first dose reduction is 30 mg once daily, and the second dose reduction is 15 mg once daily.
•  No dose adjustment is recommended for patients with mild to moderate renal impairment.
• No recommended dose has been established for severe hepatic impairment.

How to take Dacomatinib 45 mg?

• Take a 45 mg Daconib tablet orally once daily with or without food at the same time each day. Take the medication exactly as prescribed by your doctor.
• After vomiting a dose or for a missed dose, do not take an extra dose. Instead, skip that dose and take the next regular dose at the same time.
• Avoid taking certain other medications at the same time, such as proton-pump inhibitors. If you are using H2-receptor antagonists, take this at least 6 hours before and 10 hours after taking H2 antagonists.
• Consistency is important; taking the medication at the same time every day helps maintain a steady drug level.

What are the side effects of Dacomatinib?

• Severe or Fatal interstitial lung disease (ILD) can occur during the treatment. Monitor patients for pulmonary symptoms, and if confirmed, discontinue the treatment.
• Severe diarrhea can occur in patients treated with Daconib; withhold treatment or reduce the dose depending on the severity of diarrhea.
• Allergies and skin rash can occur during treatment. If grade-3 or grade-4 allergies occur, withhold the treatment until recovery, then resume the treatment with the same or reduced dose depending on severity.
• Severe embryo-fetal toxicity can occur during the treatment. It is not recommended to use Daconib in pregnancy and lactation. 

What should I tell my doctor before taking Dacomatinib?

• Tell your doctor before taking Daconib 
• If you have frequent diarrhea.
• If you have a history of breathing or lung problems other than lung cancer.
• If you are pregnant or plan to become pregnant. Daconib can harm your unborn baby.
• If you are breastfeeding or plan to breastfeed. 
• If you are taking any medications, supplements, or herbal medicine.
• Tell your healthcare provider if you have any other medical conditions or major surgery.
